British pharmaceutical giant GSK announced on Friday that it would discontinue development of camlipixant, an experimental chronic cough treatment it acquired through a two-billion-dollar deal three years ago.

The drug discontinuation followed mixed Phase 3 data, with the company conceding that the medication had failed to meet nearly all primary endpoints in two late-stage clinical trials.

GSK emphasized that the limited efficacy demonstrated was unlikely to transform patient care in any meaningful way.

GSK had originally gained access to camlipixant upon acquiring Bellus Health, a move that came despite the drug having initially failed in a Phase 2 trial.

A subsequent mid-stage study had delivered more promising results, showing a statistically significant reduction in cough frequency.

The lower dose of fifty milligrams taken twice daily, which had proved effective in that earlier study, was the highest dose evaluated in the Phase 3 clinical trials.

The termination effectively closes what had been a blockbuster opportunity for GSK, which had hoped to incorporate camlipixant into its existing portfolio of respiratory drugs, including asthma and chronic obstructive pulmonary disease treatments such as Trelegy and Anoro.

Nevertheless, the company confirmed that it is continuing Phase 2 trials of camlipixant in patients with irritable bowel syndrome, suggesting the drug may yet find a viable therapeutic application.

Camlipixant is now the third drug in the P2X3 antagonist class to fail in pharma development, following similar setbacks encountered by Bayer and Merck and Company.

Merck had reportedly spent more than five hundred million dollars to gain control of its own candidate in this class.

Industry analysts have cautioned that these repeated failures raise fundamental questions about whether P2X3 antagonists can deliver sufficient clinical benefit for chronic cough patients.

The setback does, however, clear away a competitive threat to Trevi Therapeutics, which is advancing a fundamentally different type of treatment for chronic cough.

Had camlipixant succeeded, it would have posed a significant challenge to Trevi's market prospects.

Investors will undoubtedly be scrutinizing GSK's broader pipeline strategy in the coming weeks, particularly given the substantial financial commitment the company made to secure this now-abandoned asset.
